The Key of Destruction is one of the 'minor' keys in the Pandemonium Event. It drops rarely from Nihlathak in the Halls of Vaught on Hell difficulty.
It is required for the crafting of portals to Forgotten Sands, Furnace of Pain and Matron's Den.
The other keys required are Key of Hate and Key of Terror.
